This study investigates the structural basis of poxvirus fusion regulation and the neutralization potential of antibodies targeting the A16/G9 complex. Findings indicate that A16/G9 can elicit protective immune responses in mice against vaccinia virus (VACV), positioning it as a candidate for subunit vaccines. The research highlights the urgent need for novel antiviral strategies against monkeypox, especially given its recent classification as a public health emergency. The structural insights gained may facilitate the development of effective therapeutics targeting the viral entry-fusion complex.
