This research elucidates the role of long interspersed nuclear element-1 (LINE1) in vascular calcification (VC), demonstrating that its inhibition by nucleoside reverse transcriptase inhibitors (NRTIs) significantly mitigates VC in both animal models and a clinical cohort. The study reveals that LINE1 is upregulated in calcified arteries and that its suppression downregulates the cGAS-STING signaling pathway, reducing inflammation associated with VC. Notably, a clinical study involving 1785 participants indicates that NRTIs users have a lower incidence of VC, suggesting their potential as therapeutic agents in managing this condition.
