Amlodipine, a calcium channel blocker, exhibits significant inhibitory effects on glioma stem cells (GSCs) by degrading EGFR and down-regulating pro-survival pathways. The study reveals that amlodipine induces apoptosis, reduces cell viability, and suppresses self-renewal and invasiveness of GSCs. Mechanistically, amlodipine triggers EGFR internalization via lipid raft-mediated endocytosis, leading to lysosomal degradation. These findings highlight amlodipine's potential as a repurposed therapeutic agent against glioblastoma, warranting further clinical investigation.
